Abstract The changes in concentration of vitamin C, enzymatic and antioxidant activity during the ripening of two soursop selections (G1 and G2) at room temperature (22 ºC) and refrigeration (15 ºC) with an HR 85% were evaluated. The content of soluble protein, the activity of catalase (CAT), superoxide dismutase (SOD), peroxidase (POD), the concentration of vitamin C, as well as the antioxidant activity was evaluated by DPPH, ABTS and FRAP methods. The initial soluble protein concentration of the G1 and G2 selections diminished at 22 and 15 °C during ripening. Fruits stored at 22 °C showed the highest CAT activity. The maximum activity of SOD was recorded on the sixth and fourth day in fruits stored at 22 and 15 ºC, respectively. Fruits stored at 22 °C... |

The influence of different commercial surfactants on the enzymatic activity of a commercial alpha-amylase from Bacillus licheniformis (Termamyl 300 L) has been studied. As non-ionic surfactants, alkyl polyglycosides (Glucopon® 215, Glucopon® 600 and Glucopon® 650) were studied, as were fatty alcohol ethoxylates (Findet 1214N/23 and Findet 10/15), and nonyl phenol ethoxylate (Findet 9Q/21.5NF). Also, an anionic surfactant, linear alkyl benzene sulfonate (LAS) was assayed. In general, none of the non-ionic surfactants studied, except Findet 10/15, vary substantially the enzymatic activity. Findet 10/15 has the strongest hydrophobic character and reduces the enzymatic activity more significantly the greater its concentration. Regarding LAS, this surfactant... |

Carbon is acquired through photosynthetic and non-photosynthetic processes in marine algae. However, little is known about the biochemical regulation of these metabolic pathways along the thallus of seaweeds. Consequently, the objective of this study was to assess the distribution of in vivo carboxylation pathways and to relate them to the in vitro activity of ribulose 1,5-bisphosphate carboxylase/oxygenase (RUBISCO), phosphoenolpyruvate carboxykinase (PEPCK), and phosphoenolpyruvate carboxylase (PEPC) in the Phaeophyte Laminaria setchellii, the Chlorophyte Ulva lactuca, and the Rhodophyte Iridaea cordata. Chlorophyll-a levels did not vary in U. lactuca and I. cordata. However, pigment levels were significantly lower in the meristematic region of L.... |

ABSTRACT Dormancy in lettuce seeds is a process not yet fully understood. High storage temperatures can cause seed dormancy promoting physiological and enzymatic changes. The goal of this study was to investigate the influence of storage period and environments on the quality and dormancy of different lettuce cultivars. We also investigated the biochemical changes. A completely randomized experimental design was used in a 4x8x3 factorial arrangement to evaluate physiological quality on different storage periods (30, 60, 90 and 120 days) and environments (15, 25 and 35°C) of seeds from eight lettuce cultivars (Everglades, Babá de Verão, Elisa, Luiza, Grand Rapids, Hortência, Salinas 88 and Rubete). The biochemical activity was accessed by tetrazolium test... |

The enzymatic activities involved in the digestion of proteins and carbohydrates were compared among three organs of the digestive track of two Siluriformes fish species, and between two areas: a reservoir, and an area downriver of it. Our aim was to test the hypothesis that the digestive organs of species with varied feeding habits have different enzymatic activities, and that the enzymatic activity differs among seasons and environmental conditions. The iliophagous/herbivorous species Hypostomus auroguttatus Kner, 1854 had higher trypsin-like, chymotrypsin-like peptidase and β-glucosidase activity in the intestine when compared with the omnivorous species Pimelodus maculatus Lacepède, 1803, whereas the latter had more hepatic trypsin-like activity than... |

Different parameters have been followed regularly at various times of anchovy (Engraulis encrasicholus ) ripening. Concentration in NaCl and water content as well as values of water activity and pH are constant and remain quite stable during the entire study period. The relatively acid pH value and the quite low measured water activity level created good conditions for preventing microbial putrefaction at room temperature. The free amino acids and non proteinaceous nitrogen increase indicated the modifications occurring in muscular proteins. These changes resulted in an important proteolytic activity modulated by the medium conditions, especially; pH, temperature and ionic strength. The enzymatic activity slowed down regularly with time but was not... |

Abstract: The objective of this work was to evaluate the effect of phosphites in the protection of common bean (Phaseolus vulgaris) against anthracnose. Different phosphite formulations were evaluated by quantifying peroxidase and polyphenol oxidase activity, total phenols, and lignin content. The treatments consisted of sprayings, in the V4, R5, and R7 stages, with: the K, Zn, Mn, K+Mn, K+salicyclic acid, and Cu phosphites; salicylic acid; acibenzolar-S-methyl; the fungicide azoxystrobin; besides a control, without sprayings. The area under the disease progress curve was lower in plants that received applications of the K and Mn phosphites, whose values ranged between 74 and 81%, compared with the control. The K, Zn, and K+salicyilic acid phosphites were... |

Proteolytic enzymes of hepatopancreas of six species of peneids (Penaeus kerathurus, P. japonicus, P. monodon, P. stylirostris, P. merguiensis and P. vannamei ) have been studied. Some of their physiological variations have been characterised. Increase in acclimation temperature of crustaceans enhances increase of specific activities of proteases. Variations of salinity does not modify specific activities, nor quantities of trypsin, nor electrophoretic patterns of these different proteases. |

ABSTRACT: The aim of this study was to verify the activity of some digestive enzymes in four fish species with different feeding habits. Knowledge of these enzymatic activities can help us to understand the species' digestive processes. The species chosen for this study were Ctenopharyngodon idella (herbivore), Rhamdia quelen (omnivore), Leporinus obtusidens (omnivore) and Hoplias malabaricus (carnivore). The digestive tract of these species was divided into four portions to estimate enzymatic activity: stomach, anterior, mid and posterior intestine. Ctenopharyngodon idella had the highest amylase and maltase activities in all portions of the gut, followed by L. obtusidens . The highest trypsin activity was observed in all gut portions of H. malabaricus,... |

The M17 family leucine aminopeptidase (LAP) hydrolyze amino acids from the N-terminus of peptides. Many LAPs from parasitic protozoa including Plasmmodium, Trypanosoma and Leishmania, have been intensely investigated because of their crucial roles in parasite biology. In this study, a functional recombinant Toxoplasma gondii LAP (rTgLAP) was expressed in Escherichia coli and its enzymatic activity against synthetic substrates for aminopeptidase, as well as the cellular localization was determined. Our results indicated that TgLAP is a functional aminopeptidase in the cytoplasma of T. gondii. |

A rapid and sensitive method to detect the extracellular enzymatic activity of bacteria colonies grown on agar plates is described. Selective agar media supplemented with protein, starch, chitin, Tween-80, etc. are conventionally used to detect biochemical properties of bacteria. It has been experimentally demonstrated with bacteria pure cultures that fluorogenic Methylumbelliferyl (MUF)-substrates are excellent substrate analogues for normally occurring polymers. Based on MUF-substrate hydrolysis the new method provides reliable qualitative estimates of extracellular enzymatic properties of bacteria within minutes using pure cultures as well as agar plates prepared for colony counts. |

This study aimed to obtain the condition of maximum bromalein activity in different parts of pineapple plants produced in vitro, by micropropagation. The sStems and leaves of Pérola and Imperial cultivar plants were evaluated after three and eight months of in vitro cultivation in Murashige and Skoog medium without growth phytoregulator, macerated in potassium phosphate buffer at different pH values (5.7, 6.7 and 7.7). Total protein and proteolytic activity were determined in the samples after three- and eight-month cultivation periods. For both the cultivars, the best results were obtained at pH 5.7 in extraction media. Pérola cultivar, showed higher bromelain activity in the leaves cultivated in vitro for three months (0.0194U/mL) while in the Imperial... |
