Dynamique et enzymologie de la digestion chez l'huitre Crassostrea gigas ArchiMer
Boucaud, C; Lebesnerais, C; Lubet, P; Lihrmann, I.
Feeding experiments carried out on Crassostrea gigas allowed the authors to set up the stages and the timing of the movement of food throughout the digestive tract. Various activities involved in digestion (glycosidases, esterases, and peptidases) were investigated histochemically in the digestive tract in order to localize the sites of absorption and the sites of protein and carbohydrate digestion. An explicative model of the different stages of digestion is proposed.

Bravo Rodríguez,Vicente; Jurado Alameda,Encarnación; Martínez Gallegos,Juan Francisco; Reyes Requena,Antonia; García López,Ana Isabel; Sampaio Cabral,Joaquim Manuel; Fernandes,Pedro; Pina da Fonseca,Luis Joaquim.
The influence of different commercial surfactants on the enzymatic activity of a commercial alpha-amylase from Bacillus licheniformis (Termamyl 300 L) has been studied. As non-ionic surfactants, alkyl polyglycosides (Glucopon® 215, Glucopon® 600 and Glucopon® 650) were studied, as were fatty alcohol ethoxylates (Findet 1214N/23 and Findet 10/15), and nonyl phenol ethoxylate (Findet 9Q/21.5NF). Also, an anionic surfactant, linear alkyl benzene sulfonate (LAS) was assayed. In general, none of the non-ionic surfactants studied, except Findet 10/15, vary substantially the enzymatic activity. Findet 10/15 has the strongest hydrophobic character and reduces the enzymatic activity more significantly the greater its concentration. Regarding LAS, this surfactant...

Isolation and purification of bromelain from waste peel of pineapple for therapeutic application BABT
Bresolin,Iara Rocha Antunes Pereira; Bresolin,Igor Tadeu Lazzarotto; Silveira,Edgar; Tambourgi,Elias Basile; Mazzola,Priscila Gava.
The aim of this work was to isolate and purify bromelain extracted from the pineapple peel by ammonium sulfate precipitation (40-80%), followed by desalting and freeze-drying with a 75% activity recovery and 2.2 fold increased specific activity. Ion exchange chromatography on DEAE-Sepharose was able to separate the polysaccharides from the enzyme, which was recovered in the elution step, maintaining its enzymatic activity. The batch adsorption of bromelain was evaluated in terms of total protein and enzymatic activity using Langmuir and Langmuir-Freundlich models. Results showed that the process could be suitable for the recovery and purification of the enzyme, maintaining its specific activity.

CABELLO-PASINI,ALEJANDRO; ALBERTE,RANDALL S..
Carbon is acquired through photosynthetic and non-photosynthetic processes in marine algae. However, little is known about the biochemical regulation of these metabolic pathways along the thallus of seaweeds. Consequently, the objective of this study was to assess the distribution of in vivo carboxylation pathways and to relate them to the in vitro activity of ribulose 1,5-bisphosphate carboxylase/oxygenase (RUBISCO), phosphoenolpyruvate carboxykinase (PEPCK), and phosphoenolpyruvate carboxylase (PEPC) in the Phaeophyte Laminaria setchellii, the Chlorophyte Ulva lactuca, and the Rhodophyte Iridaea cordata. Chlorophyll-a levels did not vary in U. lactuca and I. cordata. However, pigment levels were significantly lower in the meristematic region of L....

Physiological and biochemical changes in lettuce seeds during storage at different temperatures Horticultura Brasileira
Catão,Hugo Cesar RM; Gomes,Luiz Antonio Augusto; Guimarães,Renato M; Fonseca,Pedro Henrique F; Caixeta,Franciele; Galvão,Alexandre G.
ABSTRACT Dormancy in lettuce seeds is a process not yet fully understood. High storage temperatures can cause seed dormancy promoting physiological and enzymatic changes. The goal of this study was to investigate the influence of storage period and environments on the quality and dormancy of different lettuce cultivars. We also investigated the biochemical changes. A completely randomized experimental design was used in a 4x8x3 factorial arrangement to evaluate physiological quality on different storage periods (30, 60, 90 and 120 days) and environments (15, 25 and 35°C) of seeds from eight lettuce cultivars (Everglades, Babá de Verão, Elisa, Luiza, Grand Rapids, Hortência, Salinas 88 and Rubete). The biochemical activity was accessed by tetrazolium test...

Influence of season, environment and feeding habits on the enzymatic activity of peptidase and β-glucosidase in the gastrointestinal tract of two Siluriformes fishes (Teleostei) Rev. Bras. Zool.
Duarte,Silvana; Paiva,Marco A. R.; Lara,Camila C.; Bemquerer,Marcelo P.; Araújo,Francisco Gerson.
The enzymatic activities involved in the digestion of proteins and carbohydrates were compared among three organs of the digestive track of two Siluriformes fish species, and between two areas: a reservoir, and an area downriver of it. Our aim was to test the hypothesis that the digestive organs of species with varied feeding habits have different enzymatic activities, and that the enzymatic activity differs among seasons and environmental conditions. The iliophagous/herbivorous species Hypostomus auroguttatus Kner, 1854 had higher trypsin-like, chymotrypsin-like peptidase and β-glucosidase activity in the intestine when compared with the omnivorous species Pimelodus maculatus Lacepède, 1803, whereas the latter had more hepatic trypsin-like activity than...

Etude de la fraction azotee soluble de l'anchois sale en cours de maturation ArchiMer
Durand, Patrick.
Different parameters have been followed regularly at various times of anchovy (Engraulis encrasicholus ) ripening. Concentration in NaCl and water content as well as values of water activity and pH are constant and remain quite stable during the entire study period. The relatively acid pH value and the quite low measured water activity level created good conditions for preventing microbial putrefaction at room temperature. The free amino acids and non proteinaceous nitrogen increase indicated the modifications occurring in muscular proteins. These changes resulted in an important proteolytic activity modulated by the medium conditions, especially; pH, temperature and ionic strength. The enzymatic activity slowed down regularly with time but was not...

Characterization of a leucine aminopeptidase from Toxoplasma gondii OAK
Jia, Honglin; Nishikawa, Yoshifumi; Luo, Yuzi; Yamagishi, Junya; Sugimoto, Chihiro; Xuan, Xuenan.
The M17 family leucine aminopeptidase (LAP) hydrolyze amino acids from the N-terminus of peptides. Many LAPs from parasitic protozoa including Plasmmodium, Trypanosoma and Leishmania, have been intensely investigated because of their crucial roles in parasite biology. In this study, a functional recombinant Toxoplasma gondii LAP (rTgLAP) was expressed in Escherichia coli and its enzymatic activity against synthetic substrates for aminopeptidase, as well as the cellular localization was determined. Our results indicated that TgLAP is a functional aminopeptidase in the cytoplasma of T. gondii.

Microbial extracellular enzyme detection on agar plates by means of fluorogenic methylum-belliferyl-substrates. ArchiMer
Kim, S; Hoppe, H.
A rapid and sensitive method to detect the extracellular enzymatic activity of bacteria colonies grown on agar plates is described. Selective agar media supplemented with protein, starch, chitin, Tween-80, etc. are conventionally used to detect biochemical properties of bacteria. It has been experimentally demonstrated with bacteria pure cultures that fluorogenic Methylumbelliferyl (MUF)-substrates are excellent substrate analogues for normally occurring polymers. Based on MUF-substrate hydrolysis the new method provides reliable qualitative estimates of extracellular enzymatic properties of bacteria within minutes using pure cultures as well as agar plates prepared for colony counts.
